Involvement of the γ Isoform of cPLA2 in the Biosynthesis of Bioactive N- Acylethanolamines
Laboratory studyOther animals
- Design
- Laboratory study
- Subjects
- Cultured cells overexpressing single cPLA2 isoforms or pairs of isoforms; purified cPLA2 gamma enzyme
- Dose used in the study
- Not applicable; no palmitoylethanolamide was given. Cells were labelled with radioactive ethanolamine
- Duration
- Not stated in the abstract
- What was measured
- Formation of radiolabelled NAPE, lysoNAPE and N-acylethanolamines in cells expressing each cPLA2 isoform; hydrolysis of NAPE and lysoNAPE by purified cPLA2 gamma.
What the authors reported
This laboratory study found:
- Only the epsilon isoform raised NAPE production on its own.
- Cells expressing both epsilon and gamma isoforms made more lysoNAPE and N-acylethanolamine.
- Purified cPLA2 gamma hydrolysed NAPE to lysoNAPE and lysoNAPE to glycerophospho-N-acylethanolamine, which glycerophosphodiesterase 1 then converted to N-acylethanolamine.
- The authors concluded that cPLA2 gamma takes part in N-acylethanolamine biosynthesis, a pathway that produces palmitoylethanolamide.
Limits of this study
A cell and enzyme study of a biosynthetic pathway; it cannot show any effect in people or animals treated in practice. Palmitoylethanolamide itself was not tested. The authors declare no conflict of interest.
