Impact of circadian rhythmicity and sleep restriction on circulating endocannabinoid (eCB) N-arachidonoylethanolamine (anandamide)

Erin C Hanlon. Psychoneuroendocrinology. 2020 Jan:111:104471.

Randomised controlled trialHumans

Design
Randomised controlled trial
Subjects
14 young adults
Dose used in the study
No palmitoylethanolamide was given; serum levels were measured
Duration
Two nights of normal (8.5 h) or restricted (4.5 h) sleep in each condition, separated by at least one month, followed by 24-hour blood sampling
What was measured
Serum anandamide, oleoylethanolamide and palmitoylethanolamide across 24 hours with identical meals, after normal and restricted sleep.

What the authors reported

This trial found:

  • Mean 24-hour anandamide was 0.697 plus or minus 0.11 pmol/ml with a biphasic profile peaking at 0200 and 1500 and lowest at 1000.
  • Oleoylethanolamide and palmitoylethanolamide followed the same pattern.
  • None of the three was changed by sleep restriction at any time of day, unlike 2-AG in earlier work.

Limits of this study

A small randomised crossover measurement study; no PEA was given. Results describe endogenous rhythms in healthy young adults only. The author reports no conflicts of interest.
