PEA prevented early BBB disruption after cerebral ischaemic/reperfusion (I/R) injury through regulation of ROCK/MLC signaling
Laboratory studyOther animals
- Design
- Laboratory study
- Subjects
- Microvascular endothelial cells under oxygen-glucose deprivation
- Dose used in the study
- PEA in culture; ROCK2 inhibition IC50 38 micromolar
- Duration
- 0 to 4 hours
- What was measured
- ROCK/MLC signalling, stress fibre formation and endothelial barrier integrity, with and without a PPAR-alpha antagonist
What the authors reported
PEA inhibited the enzyme ROCK2 and reduced early barrier disruption after oxygen-glucose deprivation. A PPAR-alpha blocker did not remove the effect, pointing to a PPAR-alpha independent mechanism.
Limits of this study
Cell culture only. Relevant to the mechanism pages because most of the literature attributes PEA's effects to PPAR-alpha; this paper reports a target outside that pathway. Authors declare no conflicts.
