Nutraceutical formulation based on a synergic combination of melatonin and palmitoylethanolamide for the management of allergic events
Laboratory studyHumansWith melatonin
This study tested palmitoylethanolamide together with melatonin. Its results cannot be assigned to palmitoylethanolamide alone. More combination studies.
- Design
- Laboratory study
- Subjects
- Human mast cell line (HMC-1.2), stimulated to release inflammatory mediators
- Dose used in the study
- A nutraceutical formulation of melatonin and palmitoylethanolamide tested at their bioaccessible concentrations after simulated intestinal digestion (bioaccessibility: palmitoylethanolamide 1.6%, melatonin 36%)
- Duration
- Not stated in the abstract
- What was measured
- COX-2 mRNA transcription and enzyme activity, histamine and beta-hexosaminidase release, and TNF-alpha and IL-6 mRNA transcription and protein production in stimulated human mast cells
What the authors reported
The formulation reduced COX-2 mRNA transcription by 30% versus stimulated controls (p<0.0001) and inhibited COX-2 enzyme activity directly (IC50 152 micrograms/mL). It also reduced histamine and beta-hexosaminidase release and lowered TNF-alpha and IL-6 mRNA transcription and protein production in stimulated cells.
Limits of this study
A cell-line study; it cannot show effect in people. The authors declare no commercial or financial conflicts of interest.
