Activation of cannabinoid type 1 receptor (CB1) modulates oligodendroglial process branching complexity in rat hippocampal cultures stimulated by olfactory ensheathing glia-conditioned medium
Laboratory studyOther animals
- Design
- Laboratory study
- Subjects
- Rat olfactory ensheathing glia cultures and hippocampal mixed cell cultures
- Dose used in the study
- No palmitoylethanolamide was added; PEA was measured in conditioned medium. Cultures were treated with URB597 or JZL184 at 10^-9 M and AM251 at 10^-6 M
- Duration
- Not stated in the abstract
- What was measured
- Endocannabinoid system markers in olfactory ensheathing glia; anandamide, 2-AG, PEA and OEA in conditioned medium; oligodendrocyte process branching by Sholl analysis; Akt and ERK phosphorylation
What the authors reported
Olfactory ensheathing glia expressed CB1, FAAH and MAGL, and released anandamide, 2-AG, PEA and OEA. Their conditioned medium increased oligodendrocyte branching, an effect blocked by a CB1 antagonist, while medium enriched with OEA or 2-AG did not change branching in premyelinating cells and reduced it in mature cells. Akt and ERK phosphorylation did not change.
Limits of this study
A rat cell-culture study; it cannot show effect in people or animals treated in practice. PEA was measured, not tested. The authors declare no commercial or financial conflicts.
