Absorption, anti-inflammatory, antioxidant, and cardioprotective impacts of a novel fasting mimetic containing spermidine, nicotinamide, palmitoylethanolamide, and oleoylethanolamide: A pilot dose-escalation study in healthy young adult men

Rhodes CH, Hong BV, Tang X, Weng CY, Kang JW, Agus JK, Lebrilla CB, Zivkovic AM. Nutr Res. 2024;132:125-135.

Randomised controlled trialHumansWith spermidine, nicotinamide, oleoylethanolamide

This study tested palmitoylethanolamide together with spermidine, nicotinamide, oleoylethanolamide. Its results cannot be assigned to palmitoylethanolamide alone. More combination studies.

Design
Randomised controlled trial
Subjects
5 healthy young men
Dose used in the study
Low, medium or high doses of a four-compound supplement, or wheat flour control, 2 hours after a standard breakfast; PEA dose not stated in the abstract
Duration
Single doses with blood samples over 4 hours
What was measured
Plasma levels of the four compounds; TNF-alpha and reactive oxygen species from macrophages incubated with the plasma; cholesterol efflux capacity

What the authors reported

Plasma PEA, oleoylethanolamide and 1-methylnicotinamide rose after the supplement in a dose-dependent way. Macrophages exposed to post-supplement plasma released less TNF-alpha and fewer reactive oxygen species.

Limits of this study

Five participants. A pharmacokinetic and laboratory-marker study, not a clinical outcome study. Four compounds given together. Registered NCT05017428.
