Palmitoylethanolamide supplementation partly modulates eccentric exercise-induced changes in the bioactive lipid profile of skeletal muscle tissue

Schouten M, Costamagna D, Ramaekers M, Bogaerts S, Van Thienen R, Peers K, Thomis M, Dalle S, Koppo K. Pflugers Arch. 2026;478:77.

Randomised controlled trialHumans

Design
Randomised controlled trial
Subjects
10 healthy men, double-blind crossover
Dose used in the study
Levagen+ 2 x 350 mg a day or maltodextrin placebo
Duration
Each condition covered one bout of muscle-damaging exercise (240 eccentric knee-extensor contractions) with biopsies at baseline and 48 hours
What was measured
Bioactive lipid mediators in muscle biopsies by LC-MS/MS lipidomics

What the authors reported

Many lipid mediators derived from arachidonic, docosahexaenoic, linoleic and alpha-linolenic acids fell 48 hours after exercise on placebo but stayed level or rose on PEA. MCTR3 was higher and LXB4 and AT-RvD3 lower on PEA regardless of exercise.

Limits of this study

10 participants. Mechanistic outcome (muscle lipid profile), not soreness or performance. Abstract only; funding and product supply not stated in the text supplied. Same group as the 2024 Schouten trial.
